Fast and simultaneous analysis of phenylalanine and tyrosine in plasma using pillar array columns with a gradient elution system

Abstract

A rapid and quantitative method for the determination of phenylalanine (Phe) and tyrosine (Tyr) in plasma using pressure-driven liquid chromatography (LC) on a chip was developed. After derivatizing Phe and Tyr with 4-fluoro-7-nitro-2,1,3-benzoxadiazole (NBD-F), separation was performed within 140 s on a previously developed integration chip with a pillar array column and a gradient elution system [1,2]. The Phe and Tyr in plasma were quantified and good validation data were obtained. As Phe and Tyr are considered to be important biomarkers of some diseases, especially for newborn screening, the developed method is promising for rapid diagnosis in the clinical field.
