High-throughput single-cell secretion measurement on an optical wavegude chip

    We report a newly developed platform that enables us to monitor protein secretion from more than hundreds individual single cells in real time simultaneously. The platform was achieved after improvement of the limited size of view field in total internal reflection fluorescence microscopy, which was owing to using optical waveguide technology. Although the coupling of PDMS microwells and glass waveguides raised another problem regarding elevated background, it was successfully solved by sandwiched a thin layer of the amorphous fluoropolymer between the glass waveguide and the PDMS microwell. Protein secretion from about 900 cells was successfully observed on the platform.
